Zinkgluconat ist eine chemische Verbindung und stellt das Zinksalz der Gluconsäure dar.

Zinkgluconat kann durch Reaktion von Gluconsäure mit Zinkoxid und Zinkacetat gewonnen werden.[3]

Zinkgluconat ist ein weißer geruchloser Feststoff, der löslich in Wasser ist.[1] Es kommt in mehreren Hydratformen bis zum Trihydrat vor.[4]

Als Zinkverbindung wird Zinkgluconat zur Vorbeugung gegen oder Behandlung von Zinkmangelzuständen verwendet. Die WHO empfiehlt eine Zinksupplementierung als ergänzende Maßnahme in der Behandlung einer durch Durchfall entstandenen Dehydratation mit oraler Rehydrationslösung (ORS).[5] Gesundheitsbezogen werden Zinkverbindungen wie Zinkgluconat auch in der Vorbeugung gegen oder Behandlung von Erkältungskrankheiten verwendet. Für Zink in Nahrungsergänzungsmitteln hat die Europäische Behörde für Lebensmittelsicherheit (EFSA) die gesundheitsbezogene Angabe „trägt zu einer normalen Funktion des Immunsystems (körperliche Abwehr) bei“ genehmigt.

In zwei Placebo-kontrollierten Doppelblindstudien konnte gezeigt werden, dass eine Einnahme von Zinkgluconat-Lutschtabletten die Schwere von Erkältungssymptomen um ca. 40 Prozent vermindert und ihre Dauer um drei bis vier Tage verkürzt, was jedoch mit verschiedenen Nebenwirkungen verbunden sein kann. Dabei wurde in der Sherif-B.-Mossad-Studie während der Wachzeit alle zwei Stunden Zinkgluconat-Lutschtabletten mit 13,3 mg Zinkgehalt eingenommen, was einer täglichen Einnahme von max. 7–8 Lutschtabletten entsprach. Der empfohlene Tolerable Upper Intake Level der Europäischen Behörde für Lebensmittelsicherheit liegt bei 25 mg Zink pro Tag; wegen eventueller Immunschwächung vermied die Studie Zinkdosen größer als 150 mg pro Tag.[6][7][8]

Das indische Institute of Medical Education and Research konnte bei einer für die Cochrane Collaboration durchgeführten Auswertung (Metaanalyse) von insgesamt 15 publizierten klinischen Studien zur Behandlung bzw. Prävention von Erkältungskrankheiten mit Zink eine mildernde und die Krankheitsdauer verkürzende Wirkung feststellen.[9][10] Laut einer 1998 veröffentlichten Studie wies Zinkgluconat in Form von Lutschpastillen keine signifikanten positiven Effekte hinsichtlich der Erkältungsdauer bei Kindern und Jugendlichen auf. Bei Erwachsenen berichteten fünf von zehn Studien über positive Wirkungen einer Zinkgabe bei Erkältungen, einem schnelleren Abklingen der Krankheitssymptome. In den anderen fünf Studien wurde hingegen keine Wirksamkeit festgestellt.[11] In einer kontrollierten Studie hatte intranasal verabreichtes Zinkglukonat keinen Einfluss auf Schwere oder Dauer von Erkältungssymptomen, die im Zuge von Infektionen mit Rhinoviren auftraten. Nach der intranasalen Anwendung von Zinkgluconat wurde in einzelnen Fällen ein bleibender Verlust des Geruchssinns (Anosmie) berichtet.[12]

Zinc gluconate is a zinc salt of gluconic acid comprised of two gluconic acid molecules for each zinc cation (2+). Zinc gluconate is a generally recognized as safe (GRAS) substance by FDA 9. It is available as a trace mineral supplement and over the counter as a lozenge form for a reduced duration of common colds and with decreased symptom severity.

Although it has been nasally administered for treating the common cold, this route of administration has been associated with some cases of anosmia 5, 1, 2, 7.

Studies show that zinc may be better absorbed in humans in the gluconate form 4, 19, however, results from other studies may vary 10, [L27280].

Interestingly, zinc supplementation has become a critical intervention for treating diarrheal episodes in children. Studies suggest that administration of zinc along with new low osmolarity oral rehydration solutions/salts (oral rehydration solution), may reduce both the duration and severity of diarrheal episodes for up to 12 weeks 8.

Zinc gluconate is mainly indicated in conditions like zinc deficiency, and can also be administered in adjunctive therapy as an alternative drug of choice in diarrhea 15.

Although the mechanism of action is not completely known, zinc supplementation may be used to increase immunity against viruses or may interfere with the replication of certain viruses, such as the human papillomavirus (HPV) 7.

Nausea, vomiting, diarrhea, and mouth irritation have been reported in patients ingesting zinc gluconate tablets that were dissolved in the mouth for management the common cold 11.

Zinc is a naturally occurring mineral. Zinc is important for growth and for the development and health of body tissues.

Zinc gluconate is used to treat and to prevent zinc deficiency.

Before using zinc gluconate, talk to your doctor, pharmacist, herbalist, or other healthcare provider. You may not be able to use zinc gluconate if you have certain medical conditions.

Avoid taking this medication with foods that are high in calcium or phosphorus, which can make it harder for your body to absorb zinc gluconate. Foods high in calcium or phosphorus include milk, cheese, yogurt, ice cream, dried beans or peas, lentils, nuts, peanut butter, beer, cola soft drinks, and hot cocoa.

Zinc gluconate can make certain antibiotics less effective. Tell your doctor about all other medications you are using before you start taking zinc gluconate.

Before using zinc gluconate, talk to your doctor, pharmacist, herbalist, or other healthcare provider. You may not be able to use zinc gluconate if you have certain medical conditions.

It is not known whether zinc gluconate will harm an unborn baby. Do not take zinc gluconate without telling your doctor if you are pregnant or could become pregnant during treatment.

It is not known whether zinc gluconate passes into breast milk or if it could harm a nursing baby. Do not use this medication without telling your doctor if you are breast-feeding a baby.

Use exactly as directed on the label, or as prescribed by your doctor. Do not use in larger or smaller amounts or for longer than recommended.

Take zinc gluconate with a full glass of water.

Take zinc gluconate with food if it upsets your stomach.

Your healthcare provider may occasionally change your dose to make sure you get the best results from zinc gluconate. The recommended dietary allowance of zinc gluconate increases with age. Follow your healthcare provider’s instructions. You may also consult the National Academy of Sciences “Dietary Reference Intake” or the U.S. Department of Agriculture’s “Dietary Reference Intake” (formerly “Recommended Daily Allowances” or RDA) listings for more information.

Store at room temperature away from moisture and heat.

Skip the missed dose if it is almost time for your next scheduled dose. Do not use extra medicine to make up the missed dose.

Seek emergency medical attention or call the Poison Help line at 1-800-222-1222.

Overdose symptoms may include nausea, severe vomiting, dehydration, and restlessness.

Avoid taking this medication with foods that are high in calcium or phosphorus, which can make it harder for your body to absorb zinc gluconate. Foods high in calcium or phosphorus include milk, cheese, yogurt, ice cream, dried beans or peas, lentils, nuts, peanut butter, beer, cola soft drinks, and hot cocoa.

Get emergency medical help if you have any of these signs of an allergic reaction: hives; difficulty breathing; swelling of your face, lips, tongue, or throat.

Less serious side effects may include:

upset stomach.

Usual Adult Dose for Dietary Supplement:

Dosage: 105 mg to 350 mg daily.

The following drugs can interact with or be made less effective by zinc gluconate. Tell your doctor if you are using any of these:

a blood thinner such as warfarin (Coumadin);

methyltestosterone (Android, Methitest, Oreton);

penicillamine (Cuprimine, Depen);

risedronate (Actonel);

a tetracycline antibiotic such as demeclocycline (Declomycin), doxycycline (Adoxa, Doryx, Oracea, Vibramycin), minocycline (Dynacin, Minocin, Solodyn, Vectrin), or tetracycline (Brodspec, Panmycin, Sumycin, Tetracap); or

an antibiotic such as ciprofloxacin (Cipro), ofloxacin (Floxin), norfloxacin (Noroxin), levofloxacin (Levaquin), and others.

This list is not complete and other drugs may interact with zinc gluconate. Tell your healthcare provider about all medications you use. This includes prescription, over-the-counter, vitamin, and herbal products. Do not start a new medication without telling your doctor.
